Looking at one of these I would like a manual, the person who will spend most time in it is adamant that it has to be an auto. Does the tiptronic or whatever their version is called help to make it any more fun?

No. The six speed manual with the 320 engine turns the SLK from a posey tourer into a sports car. With the autobox it's not terrible because you can still select ratios, but it.s not nearly as sporty to drive.

YES.
MB manual boxes are "average-to-ordinary".
Their autos (latest with tiptronic-style - albeit it "side-to-side" as opposed to "fore-aft") are the best.
Do NOT think about MB with manual - you might like it, but buyer from you will NOT.

Agree completely.
MB manual boxes are not up to the same standard as the autos.
Expect to pay at least £2K less for a manual SLK 320 than the auto.
BTW. If you're interested in a pristine SLK 320 (auto, leather, Xenon etc. etc.) email me.
