Is no claims bonus affected by who is the named driver and who is the policy holder?

I am currently the policyholder and main driver for our car. My wife is the named driver. I/we have more than 10 years no claims bonus.

If my wife becomes the main driver will that affect the NCB? I'm concerned that she would have to start again.

Same question applies if my wife becomes the policyholder.

The situation with your no claims bonus depends on your insurer. If they have provided a certificate of no claims bonus for both you and your wife each year and you stay with the same insurer, then it should make no difference who is the policy holder and named driver. However, the majority of insurers only credit the policy holder with no claims bonus.
