Can listening to the radio cause a flat battery?

Does playing the radio with engine switched off cause battery to go flat within 20mins?

Yes, using the radio in your car with the engine switched off will drain the battery, although how long this will take depends on the vehicle. Also many vehicles will warn you if the battery is being drained excessively, and some will even automatically switch the radio off to prevent this.
